COMPARATIVE EVALUATION OF ANTIBACTERIAL EFFEICACY OF PROBIOTICS, CALCIUM HYDROXIDE AND TRIPLE ANTIBIOTIC PASTE AGAINST E.FAECALIS BIOFILM: IN VITRO STUDY

Abstract :

AIM To find whether probiotic lactobacilli have inhibition against enterococcus faecalis & compare its inhibition efficacy with triple antibiotic paste and calcium hydroxide. METHODOLOGY Agar diffusion assay is performed to identify the antibacterial efficacy. Agar plates were incubated with organisms and the test specimens that is triple antibiotic paste, calcium hydroxide, probiotic at varying concentration is noted by the zone of inhibition. The second stage was to evaluate the inhibition on biofilm formation. Higher concentrations than zone of inhibition was used to evaluate the inhibition against e.faecalis. MTT assay was performed to evaluate this. RESULTS At the minimum concentration of 250 (µg/l) the probiotic has an inhibitory range of 6.38mm and it gradually increased to 12.88mm and 39.75 at 500 and 1000(µg/l) respectively. The inhibitory action of calcium hydroxide reaches maximum of 4.75 at 1000(µg/l). Triple antibiotic has the zone of inhibition of more than 30mm at all concentrations. The viability of bacteria was higher than calcium hydroxide in probiotic group at a concentration of 1000(µg/l). When calcium hydroxide reduced up to 20% of bacteria the probiotic reduced only 8% and triple antibiotic reduced 70%of bacteria. At a concentration of 1500 and 2000(µg/l) probiotics reduced the count of bacteria 30 and 60% respectively. CONCLUSION In the coming future probiotics will be a promising replacement in the field of intracanal medicaments. Newer probiotic groups have to be tested and in vitro and clinical studies are required to confirm the same.
