Comparative Analysis of Microbial and Human Amylase Activity

Abstract :

The Aspergillus niger for amylase production was isolated from the soil sample. Aspergillus niger was streaked on two Sabouraud’s dextrose agar plates and then incubated for three days. The colonies were observed. One of the plate was used as wild type and other one as mutant type. The soil sample of Aspergillus niger was used as wild type and UV radiated Aspergillus niger on SDA plate as mutated plate. Many colonies was observed in wild type plate than mutated plate. Barley was prepared wild type and mutant type Aspergillus niger was streaked on Barley medium The colonies were observed . Wild type Aspergillus niger showed more spores than mutant type Aspergillus niger .The viability was performed results indicated the same as in wild type and mutant type. Sterilized rice water was prepared wild type and mutant type Aspergillus niger was inoculated. Wild type Aspergillus niger showed more colonies than mutant type Aspergillus niger and viability results implicated the same. The fermentation test was done by continuous and batch method for starch extraction. The amylase activity of Aspergillus niger tested by glucose qualitative analysis showed more concentration of glucose was observed in continuous method than batch method. Human serum and saliva sample was tested by semi auto analyzer and low amount of amylase was noted. The comparative study showed that amylase production was higher in soil sample of Aspergillus niger and low in Human sample.
