Evaluation of blood agar for isolation of Mycobacterium tuberculosis from sputum samples.

Abstract :

 Mycobacterium tuberculosis is a very slow growing bacterium. The gold standard in the laboratory diagnosis of tuberculosis is isolation of the bacteria in media like Lowenstein Jensen (LJ) medium and middle ook oths. Blood agar is a non–selective media for wide range of pathogenic and non–pathogenic bacteria and is widely used in clinical laboratories. We attempted to evaluate the usefulness of blood agar for isolation of M.tuberculosis from sputum samples. This study was performed on 30 AFB positive sputum samples which were inoculated onto 3 media (blood agar, LJ medium with and without PNB). Out of 30 samples, 29(96%) LJ slopes and 3(10%) blood agar slopes showed growth of Mycobacteria. The Mean time to detect visible growth of Mycobacteria was 57.6 days by blood agar versus 24.5 days by Lowenstein–Jensen medium. The present study demonstrates blood agar may not be a useful media for isolation of Mycobacterium tuberculosis.
